The
aim of this study is to examine the modulation of choline metabolism by SAHA
through a combination of magnetic resonance spectroscopy and enzymatic
studies. This study confirmed previous findings of increased choline-related
metabolites following HDAC inhibition, while providing new insight into the
underlying mechanism. HDAC inhibition was associated with increases in
choline kinase and phosphatidylcholine-specific phospholipase C activities,
suggesting phosphocholine levels are elevated as a result of upregulation in
both synthesis and catabolism.
